Yes, absolutely. But 350 twins have a long and illustrious (?!) history, whereas 500 twins are much scarcer. Just boring it out to 350cc would improve rideability at lower revs, and the state of tune wouldn't need to be so high, improving reliability. Limiting it to 250cc is madness. Anywhere between 350 and 400cc would be the sweet spot for me.

Yeah, guilty as charged But Yamaha were still producing RD350s into the early 90s (albeit made in Brazil). Anyone who fancies a 250 would (I humbly submit) be even happier with a 350 (+). I agree that as a GP race class, 350s died out in the early 80s, whereas 250s continued until 2010. But anyone who is really into two-strokes would prefer the larger engine.
